Number and variety of experimental models increases importantly the quality of education of contemporary molecular and cellular biology. The first objective of this project is to introduce the routine manipulation, RANKL induced differentiation and genetic manipulation of osteoclast precursors cell line RAW264.7, as well as RANKL cytokine preparation and purification into theoretical and practical education of course Molecular Biology of Eukaryots (Bi7090) at Faculty of Science, Masaryk University. Scientific objectives comprises the monitoring of expression of c-Jun transcription factor during osteoclast differentiation, optimization of transfection of RAW264.7 cell line and modification of these cells either with pMT-c-Jun-CD4 or pMT-v-Jun-CD4 vector and estimate the possible effects of cellular (c-Jun) or viral (v-Jun) Jun proteins on osteoclast differentiation.
