Sip of Science: Tales of a microscopist - Adapting advanced microscopy techniques for plant biology

This talk recapitulates the efforts to implement diverse microscopytechniques widely used in mammalian research in plant systems, either on fixedor living samples. First, we look at the possibility of establishing DNA andchromatin fiber stretching techniques to look at DNA methylation and histonemodifications at particular loci, then we dive into the possibilities ofin-situ DNA damage induction in living plant protoplasts. Finally, we will lookat the possibilities of correlative (electron + light microscopy) techniques tostudy plant cellular ultrastructure and DNA replication. The presentation willfeature an honest look at what current microscopy and super-resolutiontechniques are useful for and whether the hunt for finer and finer resolutioncan bring relevant advances in our understanding of cellular architecture andphysiology.
